OECD 405 Standard Test for Acute Eye Irritation, Corrosion

This test guide is preferably designed for use with albino rabbits. The test substance is administered in a single dose to the conjunctival sac of one eye of each animal. The other untreated eye acts as a control. The first test uses an animal; The dose level depends on the nature of the test substance.

If a corrosive effect is not observed in the initial test, a confirmatory test should be performed, with the irritant or adverse response confirmed using two additional animals. It is recommended to do it sequentially on one animal at a time, rather than exposing two additional animals at the same time.

The duration of the observation period should be sufficient to fully assess the magnitude and reversibility of the observed effects. Eyes should be examined at 1, 24, 48, and 72 hours after test substance administration. Ocular irritation scores should be evaluated in relation to the nature and severity of the lesions and their reversibility or irreversibility.
