This European standard specifies a multi-reference method for the determination of lipophilic algae toxins in shellfish, including raw shellfish and cooked shellfish, by liquid chromatography combined with tandem mass spectrometry LC-MS/MS.
This method was validated in a three-part interlaboratory study through analysis of both naturally contaminated blue mussel homogenates and spiked blue mussel, oyster, and oyster extracts.
The detection limit for okadaic acid group toxins, azaspiracids and pectenotoxins was determined as 6 µg/kg shellfish meat and 10 µg/kg shellfish for yessotoxins.
Quantitative determination of okadaic acid (OA), pectenotoxin 2 (PTX-2), azaspiracid-1 (AZA-1), and yessotoxin (YTX) can be performed directly by means of commercially available standard substances. Assuming an equal response factor, okadaic acid is used for the indirect quantitative determination of the two dinophysistoxins dinophysistoxin-1 (DTX-1) and dinophysistoxin 2 (DTX-2).
The limit of quantification (LOQ) for okadaic acid group toxins, azaspiracids and pectenotoxins was determined as 20 µg/kg shellfish meat and 35 µg/kg shellfish meat for yessotoxins. By hydrolysis, esters of okadaic acid, DTX-1 and DTX-2 can also be quantitatively determined as the corresponding free acids.
