The method was validated with wheat, wheat flour and wheat cracker samples. Wheat and wheat flour was prepared from a mixture of wheat and fungus-infested wheat grains. Wheat crackers were baked from wheat flour and water with added target mycotoxins.

Trichothecenes and zearalenone are extracted from the homogenized sample material with a mixture of acetonitrile and water. The extract is filtered and evaporated to dryness. The residue is dissolved with one.

Mycotoxins are purified and concentrated on the column, then liberated using methanol as eluent. Isotopically labeled mycotoxins are added to the column eluate before evaporation to dryness. After reconstitution of the dry extract with the injection solvent, mycotoxins are detected by reverse phase HPLC-MS/MS.
