ASTM E1053 Standard Test Method for the Efficacy of Virucidal Agents for Inanimate Environmental Surfaces

The ASTM E1053 test is used to evaluate the virucidal effectiveness of disinfectant solutions or pressurized disinfectant spray products on inanimate non-porous environmental surfaces. This test method is designed to be performed by persons trained in virology.

This test method can be used with most viruses and is designed for embryonated eggs or tissue culture host systems. This test method should only be performed by persons trained in microbiological or virological techniques.

If requested by the manufacturer, the test virus is loaded with organic soil to the appropriate level. This is typically done when the test product is intended to be marketed as a single-stage disinfectant. EPA requires a 5% organic soil load for a one-step disinfectant request.

The test product is prepared with the use-dilution specified by the manufacturer, if any. An equal volume of buffered saline or cell culture medium is prepared to serve as a virus recovery control. The prepared viral vaccine is spread over the entire surface of a glass Petri dish carrier and left to dry. At the end of the drying period, the test product is applied to the viral film.

Typically 2.0 ml of use-dilution test product will be added. If spray product is used, it will be applied as 6 sprays from 8 to 4 inches apart or according to Sponsor instructions as appropriate.

Upon closing of the working contact time, the test and recovery suspensions are collected using a cell scraper and neutralized by the method most suitable for the active ingredient(s) present in the test article.

Common neutralization methods include dilution in chemical neutralizers, filtration through a physical gel matrix (for example, Sephacryl gel columns), or a combination thereof.

The data generated will conform to US EPA guidelines for efficacy data required for disinfectant claims. Dried viral movies pose a significant challenge, providing "worst-case" scenario data. The test method can be modified to accommodate different types of test product application (spray, wipe, use-dilution) and different usage conditions (dirty surfaces, varying temperatures and humidity).
