Considerations are provided for sample preparation, image acquisition parameter selection, post-processing, and data interpretation.
The information in this standard is designed to apply to products containing a porous scaffold component and designed for tissue engineering repair strategies. Scaffolds can be produced from synthetic polymers (eg, absorbable polyesters) or natural materials (eg calcium phosphates), mammalian or human-derived materials (eg demineralized bone), or combinations thereof. While some considerations are provided for imaging medium to high radio-intensity materials, specific guidelines are not provided for imaging metallic scaffolds.
The applicability of these guidelines will depend on the type of scaffolding material and, as appropriate, the user's application (eg experimental design, fabricated characterization).
The microCT guidelines discussed here are best suited for in vitro sample screening. Imaging parameters may vary depending on implantation site, animal size, breathing, etc. Specific guidelines for direct in vivo imaging of scaffolds are not included because While certain imaging parameters and processing recommendations discussed in Bouxsein et al are specific to bone imaging, many of the issues and precautions to consider also apply to in vivo scaffold imaging.
