The index of refraction of a medium with respect to air is equal to the ratio of the sine of the indiens angle of a light beam in air to the sine of the angle of refraction of the refracted ray in the given medium.

Unless otherwise stated, the refractive index is measured at 589.3 ± 20 °C with reference to the wavelength of the sodium D-line (λ = 0.5 nm).

Refractometers normally determine the critical angle. In such an apparatus, the essential part is a prism with a known refractive index in contact with the liquid to be examined.

The instrument is calibrated using certified reference materials.

When using white light, the refractometer is provided with a compensation system. the instrument gives accurate readings to at least the third decimal place and is equipped with a working tool at the prescribed temperature. The thermometer is rated at 0,5°C intervals or less.

Optical rotation is the property exhibited by chiral substances to rotate the polarization plane of polarized light.

Optical rotation is considered positive (+) for left-handed items and negative (-) for left-handed items.
