Liquid chromatography is a technique used to separate a sample into its individual parts. This separation takes place based on the chemical or physical interactions of the sample with the mobile and stationary phases.
Because there are many combinations of stationary and mobile phases that can be used when separating a mixture, there are several different types of chromatography classified according to the physical states of these phases. Liquid-solid column chromatography, the most popular chromatography technique, has a liquid mobile phase that brings with it components that are slowly filtered out of the solid stationary phase.
It relies on pumps to pass a pressurized liquid solvent containing the sample mixture through a column filled with a solid adsorbent material. Each component in the sample interacts slightly differently with the adsorbent material, causing different transport rates for different components and causing the components to separate as they flow out of the column.
High Performance Liquid Chromatography
Many different chromatography techniques and systems are available for a wide variety of applications, defined as High Performance Liquid Chromatography (HPLC).
HPLC analysis focuses on macromolecule isolation via chemical interaction, affinity or hydrodynamic volume. SEC-HPLC chromatography columns work through physical interaction with the porous medium - this is a notable difference between SEC and many other liquid chromatography techniques. Chemical interaction of the sample with the column is not necessary or desirable as separation must be based solely on molecular size (relative to the Stokes radius of a particle). SEC is primarily used for the analysis of large molecules such as proteins, polymers and polysaccharides.
