The bacterial endotoxin test is used to detect or quantify endotoxins from gram-negative bacteria using amipocyte lysate (polyphemus or Tachypleus tridentatus) obtained from the horseshoe crab (Limulus).
3 technical routes are used for this test, the gel-clot technique based on gel formation; turbidimetric technique based on the development of color after cleavage of a synthetic peptide-chromogen complex.

Method A - Gel-clot method: limit test
Method B. Gel-clot method: semi-quantitative test
Method C. Turbidimetric kinetic method
Method D. Chromogenic kinetic method
Method E. Chromogenic endpoint method
Method F. Turbidimetric endpoint method
Any of the 6 methods is used for testing. In case of doubt or disagreement, the final decision is made according to Method A, unless otherwise stated in the monograph. The test is performed in a way that prevents endotoxin contamination.
