Based on the reconstructed human epidermis (RhE), it closely mimics the biochemical and physiological properties of the upper parts of the human skin in its overall design. Cell viability is measured by enzymatic conversion of the vital dye MTT to a blue formazan salt, which is measured quantitatively after extraction from tissues.

Irritant test chemicals are defined by their ability to reduce cell viability below defined threshold levels (less than or equal to 2% for UN GHS Category 50). This test guide also includes a set of performance standards for evaluating similar and modified RhE-based test methods. There are four approved test methods that follow this test guideline.

Depending on the regulatory framework and the classification system in use, this procedure can be used as an independent substitution test for an in vivo skin irritation test or as a partial substitution test within a cascade test to determine skin irritation of test substances.

The OECD guidelines for testing chemicals are a collection of approximately 150 of the most relevant internationally recognized test methods used by government, industry and independent laboratories to identify and characterize the potential hazards of chemicals.
