The EN ISO 7932 standard, developed by the International Organization for Standardization (ISO), specifies a horizontal method for numbering live Bacillus cereus with the colony count technique at 30 °C. It applies to products designed for human consumption and animal feeding, and to environmental samples in food production and food handling.
To have a viable test method, the confirmatory step was limited to MYP agar and the typical direction of hemolysis testing. Therefore, the term hypothetical was introduced to acknowledge the fact that the confirmatory stage did not distinguish B. cereus from other closely related but less encountered Bacillus species such as B. anthracis, B. thuringiensis, B. weihenstephanensis, B. mycoides.
Bacillus cereus: means a microorganism that forms colonies on the surface of a selective culture medium and produces positive confirmatory reactions under the conditions specified in this International Standard.
In the case of solid selective culture media in Petri dishes, the surface coverage of a certain amount of test sample if the initial product is liquid or, in the case of other products, a specific amount of initial suspension should be specified.
Other plates may need to be prepared under the same conditions using decimal dilutions of the test sample or the initial suspension.
The plates should be incubated aerobically for 30 hours to 18 hours at 48 °C. Calculation of the number of B. cereus per sample or per milliliter from the number of confirmed colonies obtained on the plates at the dilution levels chosen to give a significant result and validation according to the specified test should be done carefully.
