EN ISO 9308-2 specifies a method for enumerating E. coli and coliform bacteria in water. The method is based on growth of target organisms in a liquid medium and calculation of the Most Likely Number (MPN) of organisms with reference to MPN tables.
This method is applicable to all types of water, including those with significant amounts of suspended matter and those with high background heterotrophic bacteria. However, it should not be used for coliform bacteria enumeration in seawater. When used for E. coli enumeration in marine waters, a dilution of 1→10 in sterile water is usually required, although the method has been shown to work well in some marine waters with lower than normal salt concentrations.
This method relies on the detection of E. coli based on the expression of the enzyme bD-glucuronidase and as a result does not detect most enterohaemorrhagic strains of E. coli that do not typically express this enzyme. In addition, there are few other strains of E. coli that do not express bD-glucuronidase.
The selection of tests used to detect and confirm the coliform group of bacteria, including E. coli, can be considered as part of a continuous array. The extent of verification with a particular sample depends partly on the nature of the water and partly on the reasons for the inspection.
After incubation, sample wells with a yellow color equal to or more intense than that of the comparison wells are considered positive for coliform bacteria. Yellow wells that also show any degree of fluorescence are considered positive for E. coli.
