Adult worms are exposed to a standard artificial soil treated by spraying or mixing, depending on the usual method of application of the substance. Adults surviving at 14 days are weighed and the reduction in body weight reported as EC50.

The initial screening test (filter paper contact test) involves exposing worms to testing substances on moist filter paper to identify chemicals potentially toxic to earthworms in the soil. Five or more treatment levels in a geometric series and at least ten replicates (one worm per vial) for each treatment should be used.

The tests are done in the dark and for 48 hours. Artificial soil testing provides toxicity data that better represents the natural exposure of earthworms to chemicals. It involves keeping worms in a strictly defined artificial soil samples.

Five concentrations of the test substance in a geometric sequence were applied. A non-fatal concentration and a total fatal concentration should be used. Four repetitions are recommended for each treatment.
