They are a set of tools for professionals used primarily in regulatory safety testing and then in chemical and chemical product declaration, chemical registration and chemical evaluation. They can also be used for selection and sequencing of candidate chemicals during the development of new chemicals and products and in toxicology research. This set of tests covers health effects.

The method allows an LD50 to be estimated with a confidence interval, and the results allow a substance to be classified for acute toxicity according to the globally harmonized classification and labeling system of chemicals.

It is easiest to apply to materials that produce death within a few days. This test guide is designed for use with rodents (preferably female rat). Limit testing can be used efficiently to identify chemicals that are likely to have low toxicity. The test substance is administered as a single dose, usually by gavage, to fasted animals prior to dosing.

Single animals are dosed sequentially, usually at 48 hour intervals. The first animal is dosed one step below the best preliminary estimates of LD50. The second animal receives a lower dose (if the first animal dies) or a higher dose (if the first animal survives). The animals are observed with special attention for the first 4 hours, then every other day for a total of 14 days.

Weights Animals should be determined at least weekly. All animals should undergo major autopsy. Globally, the LD50 is calculated using the maximum likelihood method.
