ISO 13366 applies to the enumeration of somatic cells in cow's milk provided that the ultimately specified preconditions are met.

This method is suitable for determining reference method values ​​required for the preparation of standard test samples and for calibrating mechanized and automated cell counting methods.

A test portion of the milk to be examined is spread on a slide to form a smear. The smear is dried. During this process, cells are stained. Next, the stained cells are counted using a microscope. The number of cells counted in a defined area is multiplied by a working factor to give the number of cells per milliliter.

Unless otherwise specified, use only reagents considered analytical grade and distilled or deionized water or water of equivalent purity.

For a rectangular shape, the upper and lower inner widths on the one hand and the left and right inner heights on the other hand should not differ by more than 0,2 mm. A representative sample should have been sent to the laboratory. It must not have been damaged or altered during transport or storage.
