The ISO 13829 standard, developed by the International Standards Organization (ISO), specifies a procedure that can be used to determine the genotoxicity of water and wastewater using the umu-test. This assay is based on the detection of genotoxicity of a test sample that increases the expression of the umuC-gene-associated SOS repair system.

The genetically modified bacterium Salmonella typhimurium TA1535/pSK1002 serves as a test organism. The bacteria are exposed to different concentrations of the samples to be tested under controlled conditions. The test is based on the ability of genotoxic agents to induce the umuC-gene in Salmonella strain in response to genotoxic lesions in DNA.

Due to its ability to respond to different types of genotoxic lesions, only a single strain is required to detect different types of genotoxic agents. Stimulation of the umuC-gene is therefore a measure of the genotoxic potential of the sample. Since the umuC-gene is fused with the lacZ-gene for β-galactosidase, stimulation of the umuC-gene can be easily evaluated by determining the activity of β-galactosidase.

Tests conducted according to this International Standard should be performed by qualified professionals or a qualified testing laboratory. In applying this International Standard, in each case, it is necessary to determine whether and to what extent additional criteria should be established, depending on the range to be tested.

The modified um-assay has proven to be a sensitive test to determine the cytotoxicity and genotoxicity of toxins, with results comparable to literature data. Furthermore, the modified um-assay showed that cytotoxic concentrations causing >50% inhibition of dehydrogenase activity (DHA) can result in an unreliable estimation of genotoxicity.
