ISO 16649-2 Horizontal Method for Counting Beta-Glucuronidase Positive Escherichia Coli -Part 2 - Colony Count Technique at 5°C Using 4-Bromo-3-Chloro-44-Indolil Beta-D -Glucuronide

Developed by the International Organization for Standardization (ISO), this part of the ISO 16649 standard specifies a horizontal method for the enumeration of β-glucuronidase positive Escherichia coli in products intended for human consumption or animal feed. It uses a colony count technique at 44 °C on a solid medium containing a chromogenic component for the detection of the β-glucuronidase enzyme.

Due to the wide variety of food and feed products, this horizontal method may not be suitable in every detail for some products. In this case, different methods specific to these products can be used if absolutely necessary for legitimate technical reasons. However, every attempt should be made to apply this horizontal method as much as possible.

A later review of this part of ISO 16649 will take into account all available information on the extent to which this horizontal method was followed and, in the case of certain products, the reasons for deviations from it.

Harmonization of test methods cannot be immediate and International Standards and/or national standards may already exist for certain product groups that are not compatible with this horizontal method. When such standards are reviewed, it is hoped that they will be modified to conform to this part of ISO 16649 so that eventually the only remaining deviations from this horizontal method will be those necessary for well-known technical reasons.

Escherichia coli strains that do not grow at 44 °C, and especially those that are β-glucuronidase negative such as Escherichia coli O157, will not be detected.

β-glucuronidase positive Escherichia coli are bacteria that form typical blue colonies on tryptone-bile-glucuronide medium (TBX) at 44 °C under the conditions specified in this part of ISO 16649.

β-glucuronidase positive Escherichia coli count Determination of the number of colony forming units (CFU) of β-glucuronidase positive Escherichia coli per milliliter or sample when testing and calculations are performed in accordance with this part of ISO 16649.
