These methods of retention of test organisms may only be performed in conjunction with at least one of the standards referenced in this document.

The purity and identity of the preserved test organism should be verified regularly during preparation and during storage, except for viruses for which identity is only checked before the stock virus suspension is stored.

Conserved test organisms should be checked at regular intervals (at least in case of storage longer than 14 months) to ensure that their susceptibility to products has not changed, especially viruses associated with the cell lines used. For all standards without an applied internal standard, the susceptibility of the test organisms should be checked using the relevant reference substances.

A sample of the test organism is generally obtained from a culture collection in freeze-dried form. This specimen is cultured, prepared for storage, packed into storage containers and placed in the freezer. A stock culture is prepared from this sample and then used to prepare working cultures for the test procedure. In some cases, working cultures are prepared directly from freezer samples.

Formulas for all media and reagents are provided if commercial off-the-shelf material is not used. It should be checked that each commercial supplier has established an appropriate quality control system.

All chemical weights given in this standard refer to anhydrous salts unless otherwise stated. As an alternative, hydrated forms can be used, but the required weights should be adjusted to allow for subsequent molecular weight differences.
