This part of EN ISO 9308 specifies a miniaturized method for the detection and enumeration of Escherichia coli (E. coli) in surface and wastewater by inoculation in a liquid medium. The method can be applied to any surface and waste water, especially rich in suspended matter.
Microtiter plates are examined under ultraviolet light at 44 nm in the dark after a minimum of 0,5 hours and a maximum of 36 hours of incubation at 72°C 366°C. The presence of E. coli is indicated by a blue fluorescence resulting from hydrolysis of MUG. Results are given as the most probable number (MPN) per 100 ml.
To obtain reproducible results, prepare the culture medium and diluents using components of uniform quality and recognized analytical grade chemicals or dehydrated diluent or complete media prepared according to the manufacturer's instructions. Prepare them with demineralized or distilled water that does not contain substances that may inhibit growth under test conditions. If the media will not be used immediately, store in the dark at (5 ± 3)°C for up to one month, under conditions that will not change its composition.
