The method consists of extraction by shaking with dimethylformamide, then, after centrifugation, adding ferric ammonium citrate and ammonia to a portion of the supernatant liquid and determining the absorbance of the solution thus obtained spectrometrically at 525 nm.
Tannin content is determined spectrophotometrically at a specific wavelength, usually 280 nm, using a reference standard. This measures the absorbance of tannin compounds in the extract.
The standard provides guidelines for reporting tannin content in sorghum, typically in milligrams of tannic acid equivalent per gram of sample.
