OECD TG488 Transgenic Rodent Somatic and Germ Cell Gene Mutation Assays Test

This test guide describes an in vivo test that detects chemicals that can induce gene mutations in somatic and germ cells. In this assay, transgenic rats or mice containing multiple copies of chromosomally integrated plasmid or phage shuttle vectors are used.

Transgenes contain reporter genes for detection of various types of mutations induced by test chemicals. A negative control group and at least 3 transgenic animal treatment groups are treated for 28 consecutive days. Administration is followed by a period in which the agent is not administered and unrepaired DNA lesions are fixed as stable mutations prior to sacrifice.

At the end of this period (usually 3 or 28 days depending on cell type and requirements), animals are sacrificed, genomic DNA is isolated from tissues of interest and purified. Mutations that occur during treatment are scored by recovering the transgene and analyzing the phenotype of the reporter gene in a bacterial host deficient for the reporter gene.
