Developed by the International Organization for Standardization (ISO), this part of the ISO 21527 standard is a horizontal scale for counting live yeasts and molds in products with a water activity greater than 0,95 [eggs, meat, dairy products (except milk powder), human consumption or animal feed. specifies the method. fruits, vegetables, fresh dough, etc.] indicates a horizontal method performed with the colony counting technique at 25 ºC ± 1 ºC.

This part of ISO 21527 does not allow the enumeration of mold spores. Neither the identification of fungal flora nor the examination of foods for mycotoxins are within the scope of this part of ISO 21527. The method specified in this part of ISO 21527 is not suitable for enumeration of heat-resistant fungi such as Byssochlamys fulva or Byssochlamys.

Due to the wide variety of food and feed products, applications of the horizontal method specified in ISO 21527 (all parts) may not be suitable for certain products. In this case, different methods specific to these products can be used if absolutely necessary for legitimate technical reasons. However, every attempt will be made to apply the horizontal method specified in ISO 21527 (all parts) whenever possible.

The next time ISO 21527 (all parts) is reviewed, all available information on the extent to which the horizontal method is followed and, in the case of certain products, the reasons for deviations from this method will be taken into account.

Harmonization of test methods cannot be immediate and for certain product groups International Standards or national standards may already exist that do not comply with the horizontal method specified in ISO 21527 (all parts). When such standards are reviewed, they will be modified to conform to ISO 21527 (all parts).
It is hoped that eventually the only remaining deviations from this horizontal method will be those required for well-known technical reasons.

Surface grafted plates are prepared using a specific selective culture medium. Depending on the expected number of colonies, a certain amount of the sample (if the product is liquid) or an initial suspension (in the case of other products) or decimal dilutions of the sample/suspension is used.
