ISO 7954 General Guide for Microbiology, Yeast and Mold Counting, 25 Degree Colony Counting Technique Test

The method consists of preparing cast plates using a specific selective culture medium and a given amount of test sample, additional dilutions of the test sample. After 25, 3 or 4 days of aerobic incubation of the plates at 5 degrees Celsius, the number of yeasts and molds per gram or milliliter of sample is calculated from the number of colonies obtained on the plates selected at the dilution levels.

This International standard is intended to provide general guidance for the microbiological examination of food products not covered by current international standards and to be considered by organizations preparing microbiological testing methods for application to foods or animal feeds.

Due to the wide variety of products in this field of application, this guide may not be suitable in every detail for some products, while it may be necessary to use different methods for some products.

In any event, however, it is hoped that every attempt will be made to apply the guidelines provided to the fullest extent possible, and that deviations from them will only be made if absolutely necessary for technical reasons. The next time this International Standard is reviewed, all available information regarding the extent to which the guidelines have been followed and the reasons for deviating from them for certain products will be taken into account.

Harmonization of test methods cannot be immediate and International Standards or national standards may already exist for certain product groups that do not comply with these guidelines. Where International Standards already exist for the product to be tested, they should be followed, but when these standards are reviewed it is hoped that these standards will be modified to align with this International Standard so that any remaining deviations from these guidelines will eventually disappear. should be those necessary for established technical reasons.
