OECD 420 OECD Guidelines for the Testing of Chemicals, Standard Test for Acute Oral Toxicity, Fixed Dose Procedure

The OECD chemicals testing guidelines are a collection of approximately 150 of the most relevant internationally recognized test methods used by government, industry and independent laboratories to identify and characterize the potential hazards of chemicals.

They are a set of tools for professionals used primarily in regulatory safety testing and then in chemical and chemical product declaration, chemical registration and chemical evaluation. They can also be used for selection and sequencing of candidate chemicals during the development of new chemicals and products and in toxicology research. This set of tests covers health effects.

One principle of the method is to use only moderately toxic doses in the main study and avoid administration of doses expected to be lethal.

This Guide is primarily intended for use with rats. Groups of animals of one sex (normally females) are dosed in a stepwise procedure using fixed doses of 5, 50, 300 and 2000 mg/kg (exceptionally 5000 mg/kg). The starting dose level is chosen based on an observational study, as the dose expected to produce some signs of toxicity without causing serious toxic effects or death.

Other groups of animals may be dosed at higher or lower fixed doses depending on the presence or absence of signs of toxicity or death. This procedure continues until the dose causing significant toxicity or death is determined, or no effects are seen at the highest dose, or deaths occur at the lowest dose.

The test substance is administered as a single dose by gavage using a gastric tube or a suitable intubation cannula. Animals should be fasted prior to dosing. A total of five animals of one sex will normally be used for each dose level investigated.
