The test substance, normally at 20 mg C/L as the sole carbon and energy source, is incubated (normally for 28 days) in sealed bottles with aerobic conditions containing a buffer-mineral salts environment.

To control the test procedure, a reference substance with known biodegradability (aniline, sodium benzoate or ethylene glycol and 1-octanol) should be tested in parallel. It is recommended to analyze triplicate vials after a sufficient number of time intervals.

Also, at least five test vials (from test containers, empty controls, and containers containing reference substance) are analyzed at the end of the test to allow calculation of 95% confidence intervals for the mean percent biodegradation value.

The evolution of CO2 from the eventual aerobic biodegradation of the test substance is determined by measuring the Inorganic Carbon (IC) produced in test bottles, which is more than that produced in empty containers containing inoculated media only.
