The ISO 4832 standard, developed by the International Standards Organization (ISO), gives general guidelines for counting coliforms by the technique of counting colonies after incubation in a solid medium. It applies to products intended for human consumption and animal nutrition, and to environmental examples in the field of food production and food processing.
Due to the wide variety of food and feed products, this horizontal method may not be suitable in every detail for some products. In this case, different methods specific to these products can be used if absolutely necessary for legitimate technical reasons. However, every attempt should be made to apply this horizontal method as much as possible.
When ISO 4832 is reviewed later, all available information regarding the extent to which this horizontal method has been followed and, in the case of certain products, the reasons for deviations from this method will be taken into account.
Harmonization of test methods cannot be immediate and International Standards and national standards that are not compatible with this horizontal method may already exist for certain product groups. When such standards are reviewed, it is hoped that they will be modified to conform to this part of ISO 6888 so that eventually the only remaining deviations from this horizontal method will be those necessary for well-known technical reasons.
It is important to inoculate the surface of the solid selective culture medium using double plates with a certain amount of initial suspension if the product is liquid, or a certain amount of test sample in the case of other products.
The two poured plates are prepared using a solid selective culture medium with a certain amount of test sample if the first product is liquid, or a certain amount of initial suspension in the case of other products.
Other pairs of poured plates are prepared using decimal dilutions of the test sample or initial suspension under the same conditions.
