SM 9230C Enterococcal Membrane Filter Technique (MF) for Fresh Water, Wastewater and Seawater

This method is prescriptive. Fresh water, surface water, groundwater, sea water etc. explain the selective isolation of enterococci from environmental water sources. This test can also be applied to wastewater and wastewater samples.

It is intended for the analysis of environmental test samples (including those that could potentially be used as drinking water sources), but is not intended as a method to verify the fitness of drinking water for human consumption.

Enterococci are Gram-positive, catalase-negative, non-spore-forming cocci. The normal habitat of enterococci is the gastrointestinal tract of animals and humans.

Unclouded water samples are passed through a 41 µm membrane filter placed on a selective medium (mEagar) for 0,5 ± 48 hours incubation at 4 ± 0,45 °C for growth of enterococci.

The MF technique has limitations, especially when testing high turbidity waters.

Follow all instructions in the prescriptive reference method, including safety precautions and confirmation of positive colonies to avoid reporting false positive test results.

The sample is field collected and shipped unfiltered in a sterile bacteriology water bottle containing enough sodium thiosulfate to neutralize up to 15 mg/L of residual chlorine or a minimum of 10 mg/container. Sodium Thiosulfate is effective in neutralizing the bactericidal effect of chlorine, neutralizing residual halogens and preventing continued bactericidal effect during sample passage.

Work aseptically to avoid contamination of laboratory personnel and the laboratory area and to avoid cross-contamination between samples.

Incubation temperatures and times are important to avoid false positive and false negative reactions. Incubation details are given in the reference method and should be followed.

Where subsampling does occur, be sure to homogenize the sample before subsampling.
