The features of the plate counting method are:
A test sample and a control sample are inoculated with the spore suspension of reference fungi and incubated at 30°C for 48 hours.
In this part of ISO 13629, fungal growth is quantitatively determined by visual counting in CFU of colonies on the agar plate and fungal activity is calculated in CFUs.
If the test sample absorbs water, the absorption method is recommended. If the test sample does not absorb water, the transfer method is recommended.
The reagents used in the tests must be of analytical purity or suitable for microbiological purposes. Commercially available dehydrated products are recommended to be used in the preparation of the culture medium in strict accordance with the manufacturer's instructions.
