ISO 20418-1 specifies a qualitative and quantitative test method for determining the content of wool, cashmere, yak fibers and their blends in textiles by microscopy pre-scan, protein extraction, enzymatic digestion and specific peptide detection using a liquid chromatography-mass spectrometer. Equipped with an electrospray ionization source (LCI-ESI-MC).
This standard can be applied to related textile products at every process stage (ie from raw material to clothing) with homogeneous distribution of components. It can be applied to different types of textile materials (eg staples, tops, yarns and fabrics) containing wool, cashmere or yak fibers and their blends.
The method is based on the preliminary identification of all fibers in the mixture based on their morphology by light microscopy. The proteins are then extracted with a thiourea/urea/dithiothreitol (DTT) solution.
An enzymatic digestion of the protein extracted from the fibers with trypsin is carried out. Analysis of specific markers is done by LC-MS and percent composition is calculated.
This standard can be applied to samples containing other fiber types other than wool, cashmere and yak, by combining its results with those obtained using the ISO 1833 series and/or the ISO 17751 series.
ISO 20418-1 does not apply if fibers from the same animal species are present (eg blends of cashmere and mohair); in this case, quantitative analysis can be performed using microscopic analysis (eg ISO 17751 series).
