This standard met the need for a relatively quick and easy-to-apply qualitative method for determining the antibacterial activity of diffusible antimicrobial agents on treated textile materials.
The standard is to detect the bacteriostatic activity on textile materials. Results from using this procedure have been demonstrated by the RA31 Committee to be reproducible by various laboratories working with materials containing residual amounts of antibacterial agent (as determined by chemical assay) after multiple standard washes.
The standard is useful for obtaining a rough estimate of activity, as the growth of the inoculation organism decreases as growth of the inoculation organism results from one end of each array to the other and with increasing degrees of sensitivity from one line to the other. The size of the zone of inhibition and the narrowing of the lines caused by the presence of the antibacterial agent allow an estimate of the residual antibacterial activity after many washes.
The standard is a quantitative procedure that is sensitive enough but laborious and time-consuming for routine quality control and screening tests. Therefore, when the goal is to demonstrate bacteriostatic activity by diffusion of antibacterial agent through agar, AATCC TM147 fulfills this need. In the Parallel Spread Method, the agar surface is inoculated, making it easier to distinguish between the test organism and contaminating organisms that may be present on the unsterilized sample. The Parallel Line Method has proven effective over several years of use in providing evidence of antibacterial activity against both Gram positive and Gram negative bacteria.
