The International Standard EN ISO 734 specifies a method for the determination of the so-called "oil content" of hexane extract (or light petroleum extract) of meals (excluding compound products) obtained by the extraction of oil from oilseeds by pressure or solvents.

A method for determining the oil content of oilseeds is specified in ISO 659. Therefore, it is necessary to control oil production by establishing a reference method for determining the oil content of oilseed meals.
A test portion of the product is extracted in a suitable apparatus with technical hexane or, in its absence, light petroleum. The solvent is eliminated and the resulting extract is weighed. Unless otherwise stated, only accepted analytical grade reagents are used.
The mechanical grinder is used to obtain particles that are easy to clean and completely pass through a 1 mm mesh sieve, allowing food to be ground without heating and without significant change in moisture, volatile matter and oil content.
In laboratories where a micro-grinder is not available, micro-grinding of the ground sample can be replaced by grinding with a pestle and pestle in the presence of approximately 10 g of sand that has been washed with hydrochloric acid and then calcined. However, milling in a mortar cannot be applied in the case of multiple analysis, as operator fatigue prevents a large number of samples from being milled efficiently enough and oil extraction from a coarsely milled sample can never be completed.
A representative sample should have been sent to the laboratory. It must not have been damaged or altered during transport or storage. Sampling is not part of the method outlined in this International Standard. A recommended sampling method is given in ISO 5500.
