Autoclave Test

Autoclaves provide a physical method for disinfection and sterilization purposes. These vehicles work with a combination of steam, pressure and time. Autoclaves use high temperature and pressure to kill microorganisms and spores. Autoclaves are used to decontaminate certain biological wastes and sterilize instruments and labware. It is recommended that legally regulated medical wastes containing bacteria, viruses and other biological materials should be inactivated by autoclaving before disposal.

For an autoclave to be effective, it must reach a temperature of 15 degrees Celsius using saturated steam at a pressure of at least 121 psi and maintain this temperature for at least 30 minutes. Depending on the nature and volume of the load, the cycle time may need to be extended. Flammable, reactive, corrosive, toxic or radioactive substances, bleach, any liquid in a closed container, and any material that comes into contact with the inner surfaces of the autoclave should never be autoclaved. In contrast, polypropylene is an inexpensive resin that can withstand autoclave temperatures. Polypropylene containers are often used as secondary containers to hold autoclaved materials. Polycarbonate can also withstand high temperatures. Polystyrene, polyvinyl chloride (PVC), nylon, acrylic, low-density polyethylene and high-density polyethylene labware and polyurethane pipes cannot be autoclaved under any circumstances.

Autoclaving, sometimes called steam sterilization, is the use of pressurized steam to kill infectious agents and denature proteins. This practice is the most reliable way to sterilize laboratory equipment and decontaminate biohazardous waste. However, autoclaves do not remove chemical contamination.

There are two basic autoclave cycles: gravity or fast exhaust and liquid or slow exhaust. The first method is used for dry goods, glassware and similar materials. This cycle fills the chamber with steam and maintains it at a certain pressure and temperature for a certain period of time. At the end of the cycle, a valve opens and the chamber quickly returns to atmospheric pressure. Drying time can also be added to the cycle. The second method prevents sterilized liquids from boiling. The steam is slowly depleted at the end of the cycle, allowing the liquids to cool.

A medical autoclave is a device that uses steam to sterilize medical equipment and other objects. This means that all bacteria, viruses, fungi and spores are inactivated. The autoclave works on the principle of moist heat sterilization. The high pressure inside the chamber raises the boiling point of the water to sterilize the equipment. The higher pressure also allows the heat to penetrate the deeper parts of the equipment quickly. The moisture in the steam causes the proteins of the microbes to coagulate, causing them to irreversibly lose their activity and function.

Autoclaves of all sizes operate on a single principle, which includes three cyclic sterilization steps:

  • Evacuation phase: The air in the sealed chamber is replaced by steam passing through the sterilizer.
  • Exposure phase: In this phase, the exhaust valve is closed and the temperature and pressure inside the sealed chamber are raised to the desired set point. The temperature is maintained for the set time.
  • Exhaust stage: The exhaust valve opens, steam is removed and the chamber is brought back to normal temperature.

The pressure chamber is the main body of the autoclave. It consists of an inner chamber and an outer sheath. Generally, the inner chamber is made of stainless steel, and the outer chamber is made of iron case.
