This standard can be considered part of a set produced by ISO/TC 126 that describes the determination of total and non-nicotine dry particulate matter (NFDPM) in total particulate matter from mainstream smoke.
A related International standard, ISO 3400, determines total alkaloids, while this standard determines only nicotine, thanks to gas-chromatographic separation. Sometimes, differences can occur in some types of tobacco due to the small amount of alkaloids other than nicotine.
The total particulate matter required to be recovered from mainstream smoke in accordance with ISO 4387 is dissolved in an extraction solution containing an internal standard. The nicotine content of a portion of the smoke extract is determined by gas chromatography and the nicotine content in the total particulate matter from mainstream smoke is calculated.
Calculate the ratio of the nicotine peak to the internal standard peak from the peak area (or height) data for each of the calibration solutions. Plot the area ratios against nicotine concentrations and calculate a linear regression equation (area ratios over nicotine concentrations) from these data. The graph must be linear and the regression line must pass through the origin.
